Le rôle fondamental de l’angiogenèse dans la croissance et la dissémination tumorale est connu depuis maintenant plus de 30 ans. Nous disposons depuis peu de médicaments anti-angiogéniques efficaces sur de nombreuses tumeurs. Leur utilisation requiert une bonne connaissance de leur mode d’action, leurs effets secondaires, leurs indications et leur place dans la stratégie thérapeutique. Cet ouvrage est donc divisé en trois points: - Il est d'abord rappelé le rôle important de l’angiogenèse tumorale, les moyens disponibles pour l’évaluer sur des prélèvements histologiques, ses modalités pratiques d’utilisation et les effets secondaires des médicaments anti-angiogéniques.- Puis les résultats thérapeutiques obtenus avec les anticorps anti-VEGF et les inhibiteurs de tyrosine kinase anti-VEGFR sont détaillés pour chacune des tumeurs.- Enfin, il est discuté le positionnement de ces nouvelles molécules dans la stratégie multidisciplinaire vis-à-vis des autres modalités de traitement.
